High Sensitivity SSC and Fluorescence Detection Using the NovoCyte Advanteon Flow Cytometer

The detection of small biological particles, such as extracellular vesicles (EVs), is critical for advancing disease research and biomarker discovery. Traditional flow cytometers often struggle with this due to limited sensitivity.

This application note demonstrates how the NovoCyte Advanteon flow cytometer, equipped with advanced detectors, delivers high-sensitivity side scatter (SSC) and fluorescence detection for superior small particle analysis.

Key Findings


Beads as small as 100 nm were clearly resolved using SSC alone.


Biological EVs (e.g., lipoparticles and platelet-derived EVs) were accurately detected and quantified, with validation against dilution and detergent controls.


Platelet-derived EVs (100–250 nm) released upon ADP stimulation were successfully identified using markers CD9, CD61, and Annexin V, with signal disappearance after detergent treatment confirming vesicle integrity.


Absolute counting enabled by the syringe pump ensures precise measurement without external beads.

Three commercially available small particle beads were used to demonstrate the high sensitivity of the NovoCyte Advanteon side scatter detection (SSC): Bang’s Laboratories NIST traceable size standards (A), ApogeeMix beads (B), and BioCytex Megamix-Plus (C).
